"The cells in the sections were stained with iron haematoxylin, a dye that provides good contrast between the chromosomes and the surrounding cytoplasm. Painter studied spermatogonial cells, both in mitosis and in the first meiotic division. Based on his analysis, he concluded "that extended study has shown that 48 is, in all probability, the correct diploid or somatic chromosome number."
Painter's conclusion was accepted for many years because the cytological techniques used during the first half of the twentieth century did not permit accurate chromosome counts, especially when large numbers of chromosomes were present. Painter's counting error was corrected in 1956 when Joe Hin Tjio and Albert Levan, working at the Institute of Genetics in Lund, Sweden, published analyses of chromosomes in dividing human embryonic cells. The previous year, Levan had tried out modifications of techniques that had been developed by T. C. Hsu for studing human chromosomes. In 1952 Hsu had reported that explants of human embryonic tissues that had accidentally been washed with a hypotonic solution yielded mitotic cells that could be analyzed easily."

Decemberists in Concert  
Tonight was alot of fun.

I want to have Colin Meloy's baby.

But really, they're fun in concert. The Venue was cool. I didn't really expect to get wanded and patted down upon entry though, that was interesting.

On the last song, they reenacted an altogether unnotable and long-forgot tragedy of the Crimean War put to poetry on the floor with some of the band members. There was also a la-di-da-di-da-didi-didi-da-ing contest between two halves of the attendees replete with leering, fistshaking, and a No-Man's Land infested with snakes.

We got lost in Philadelphia for an hour or so trying to get out. But all in all a good night.

I have cookies now too.

The Lake House  
I just watched The Lake House on DVD.

I shouldn't ever watch sappy romance movies..

I very nearly cried...

I hate myself right now >.<

But it was so beautiful.. and yeah..

Period Movies  
I just finished watching The Proposition. And Wow....

Just incredible.

Dark, violent, depressing and slow.. but incredible.

I guess I didn't mention it before.. but I saw The Illusionist a couple weeks ago at The Senator Theatre. Also an incredibly good film.

Cooking with Zach  
So I was feeling a little depressed/bored earlier. I decided to do some experimental cooking (winging it).

It turned out pretty tasty, and I thought I'd save my effort for posterity and share it with all the people who read this that may want to try it out. Because I imagine there are a fair amount of people that read my LJ that don't know how to cook at all, I'm going to be very detailed in the prep work as well as the cooking (So no one gets lost!)

I bought all the ingredients on a random run through Superfresh, and managed to spend about $20. Except for Olive Oil and garlic which I already had. The oil could up the price a bit, but I'd definitely use Olive and not regular vegetable oil or some other such thing.

AFTERMATH

If you read through this whole thing, congratulations. I hope at least someone tries it. Or visits me and cooks with me or something like that. I'm probably going to do this again, and if generally you all don't need all the gritty details, feel free to let me know so I don't have to type so much :)

The recipe is easy to multiply and could also be refined a bit. I think it turned out rather well though. I was worried about half-way through because the chicken looked a bit plain, but the sherry gave it a nice color towards the end. All told, it probably took me a little over an hour, and I was really taking my time.

If you try this and hate it, sorry. But oh well.

Half Empty?  
So...

Went to see a A Scanner Darkly tonight with Spanky.

I was quite impressed. It really articulated alot of my perspectives on reality in a way I found enjoyable. The animation technique was absolutely orgasmic. The characters are cool and the whole story was very trippy.

I liked it.

Can't really review it well right now though, too distracted.

We're thinking about maybe doing another Halo 2 LAN tomorrow night.

We'll see. Most everyone knows my cell, or comment. It would be 7 or 8pm at the earliest.
